![]() Set during Bonnie’s winter vacation, Ken ( Michael Keaton) and Barbie ( Jodi Benson) think that they’re going with her. The first “Toy Story Toon” short, released a year after Toy Story 3 (and attached to Cars 2), “ Hawaiian Vacation” is a beautifully animated charmer. “Partysaurus Rex” is notable for being primarily animated at the now shuttered Pixar Canada campus, for being the final “Toy Story Toon” (for now) and for inspiring an after-hours hard-ticket event at Walt Disney World’s water park Typhoon Lagoon. But it’s ultimately pretty one-note and Rex’s decision to ultimately flood the bathtub seems questionable. The short, written and directed by longtime Pixar artist Mark Walsh, is really, really funny and the way that Rex transforms a suburban bathtub into a debauched rave is spellbinding, with some of the most extreme and vibrant lighting that Pixar has ever done and some killer dance tunes courtesy of American electronic artist BT. Thus, Partypooper Rex is transformed into Partysaurus Rex. Dubbed “Partypooper Rex” by Potato Head, he heads off to the bathtub with Bonnie and when she gets out, is coerced by the other bathtub toys to turn the water back on and get the party going. “What up fishes?” The concept for “ Partysaurus Rex,” a short film that accompanied the 3D theatrical re-release of Finding Nemo (remember when they were doing that?), is pretty simple: the other toys label Rex ( Wallace Shawn) a buzzkill after he literally bursts a giant bubble. And having Tom Hanks’ brother Jim fill in for him as Woody (as he often does on theme park attractions, videogames and toys) doesn’t help anything either. There are definitely some cute moments, like a montage of Bo’s environments over the years, and it’s great to be back in Toy Story 4’s irresistible carnival setting at the beginning and end of the short, but this short is kind of a drag. But there isn’t a lot of story there it isn’t all that funny or interesting and feels like the equivalent of Toy Story fan fiction. "Lamp Life" was written and directed by Valerie LaPointe, a story artist on Toy Story 4 who was involved with the (re) development of the Bo character (voiced here once again by Annie Potts), so it’s understandable as to why she would want to make a short film detailing the time between when Bo left the gang and when she and Woody reconnected. ![]() Unfortunately, the short itself is pretty lackluster. Originally intended as a bonus feature for the home video release of Toy Story 4, “ Lamp Life” got some additional buzz when it became a Disney+ exclusive. But it’s more of an oddball piece of Toy Story miscellanea, than an actual compelling piece of the larger story. It’s directed by Tad Stones, the guiding force of the early days of Walt Disney Television Animation and the creator of Darkwing Duck, so the story is competently told. The rest of the movie isn’t bad, exactly. ( Tom Hanks’ brother, Jim, voices Woody.) Pixar also animated the logo and a graphics-heavy introduction of the film’s characters. Potato Head doesn’t appear due to a dispute at the time with Hasbro. Weirdly, Finding Nemo director Andrew Stanton voices Hamm and Mr. Lee Ermey, Wallace Shawn, and Joe Ranft (as Wheezy). They created a nearly three-minute opening sequence, featuring Tim Allen (who reprises his role for the rest of the movie), R. While Pixar wasn’t involved in the television series, aside from creating the wordless, 30-second intro, they were much more present for the movie/pilot. It was a traditionally animated, direct-to-video feature that also served as the extended pilot to the short-lived animated series Buzz Lightyear of Star Command. If you’ve never heard of Buzz Lightyear of Star Command: The Adventure Begins, that’s okay. The Toy Story universe is large and expansive - and we’re celebrating it all. Even if Boot Camp was an option for modern Macs, because the M-series processors are not compatible with the with the x86 version of Windows, they would only be able to run an ARM version of Windows. These new Macs that are based on the M1 chip – or any of its successors including the M1 Pro and Max, the M2 and the M2 Pro and Max – don’t offer Boot Camp. Rather than the x86 architecture used by Intel processors, the Apple’s M1 processors use a different architecture, known as ARM (named after Arm Ltd, the British company that originally designed the chips). However, in 2020 Apple announced that it would be moving away from using Intel processors to its own Apple Silicon processors and this transition began with the arrival of the ARM-based M1 Chip in November 2020. There are millions of Intel-based Macs still in use all around the world that can still use either Boot Camp or virtualization to run Windows. The only disadvantage is that you need a pretty fast Mac with plenty of memory and storage to run Windows as a VM, since it means that your Mac is effectively running two operating systems at the same time. This is a great solution as it allows you to run the Windows apps that you need right alongside your main Mac apps as well. So you can create a VM on your Mac and then install Windows – or another operating systems, such as Linux – on the VM. Virtualization programs such as Parallels Desktop and VMWare Fusion run on the macOS, but allow you to create a Virtual Machine – or VM – that mimics the hardware of a Windows PC. Luckily there are plenty, many of which use virtualization to create a Virtual Machine that emulates the computer system required to run the operating system of your choice. In the case of M-series Macs, and for anyone with an older Mac that wants to run Windows 11, or two or more operating systems side by side, you’ll need an alternative to Boot Camp. There are third-party solutions that address both of these issues. Another issue, specifically with Windows 11 on Macs, is that Windows 11 requires the presence of a TPM (trusted platform module) to run and Macs don’t have these. Many people find it inconvenient to constantly switch back and forth between the macOS and Windows. To annotate heterozygous bases, select all of the chromatograms in the Trim ends folder, click Pre-processing and select Find Heterozygotes in the dropdown. In this exercise, you will learn how to identify heterozygous bases in chromatograms. Find heterozygotes identifies heterozygotes in sequences with trace information by looking at the relative height of traces at peak positions. Heterogeneity in base sequence occurs when a single peak position within a trace contains more than one peak. If you are planning to assemble your sequences prior to running the Antibody Annotator, you can skip this section and proceed to the next section. This section explains how to find heterozygous bases in sequences without first assembling the sequences into a contig. **Note that sequence trimming is not necessary for downstream analysis such as sequence assembly and annotation. This will produce 8 documents in the Trim ends folder where the sequences are trimmed and shorter than the original input sequences. This option trims bases up until the point where trimming further bases will only improve the error rate by less than the limit (see image below). Select the Error Probability Limit option located under Trim By Quality and click Run. Trim the poor quality bases off the ends of the sequences by selecting all the chromatograms in the Input data folder, then, click Pre-processing Trim End. In this exercise, you will learn how to trim low-quality bases from both ends of chromatogram sequences. This is because the noise introduced by low-quality regions and vector contamination can produce incorrect assemblies. Trimming low-quality ends of sequences is normally performed before assembling a contig. Below is our video on Pre-processing Sanger Sequences.
